چکیده

The JAK (Janus kinase)-STAT (Signal transducer and activator of transcription) signal transduction pathway is a cascade of downstream cellular events initiated from outside of the cell through the cell surface to the DNA in the nucleus, causing transcription. The conventional modeling approach for signal transduction pathways involves solving ordinary differential equations (ODEs). We study here a computational alternative. We build two models of 46 reactions in the JAKSTAT pathway and compare the results. We implement a deterministic mathematical model using the ODEs solver COPASI, and we build a stochastic computational model using the Stochastic Pi Machine (SPiM). Since dysregulation in the functionality of JAK-STAT pathway results in immune deficiency syndrome and cancers, like lymphomas, leukemia and breast cancer, we believe that models like this have the potential to contribute to cancer research.

JAK-STAT pathway and JAK inhibitors: a primer for dermatologists

Background: All cellular events depend upon the DNA synthesis and gene expression involving complex interplay between ligands such as interleukins and interferons, with various cell membrane receptors. These ligand-receptors interactions transmit signals within the cell via numerous signal transduction pathways to affect gene expression. Janus kinase/signal transducer and activator of transcrip...

The Jak-Stat Signaling Pathway of Interferons System: Snapshots

Interferons (IFNs) are a family of small regulatory glycoproteins that play a central role in the defense against viral infections. Although IFNs have been initially discovered as antiviral factors, today they are known as an integral part of the cytokine network that affect a wide range of biological processes. IFNs exert their pleiotropic effects through their multisubunit cell surface recept...

Mathematical Model of IL-6 signal transduction in Hepatocytes

This work focuses on the role of IL-6 in the hepatic acute phase response. A kinetic model of the signal transduction network in hepatocytes stimulated by IL-6 has been developed. Signaling induced by the IL-6 family of cytokines involves the activation of two major pathways: the JAK (Janus-associated kinases)/ STAT (signal transducers and transcription factors) and the MAPK (mitogen-activated ...

Computational models of the JAK1/2-STAT1 signaling

Despite a conceptually simple mechanism of signaling, the JAK-STAT pathway exhibits considerable behavioral complexity. Computational pathway models are tools to investigate in detail signaling process. They integrate well with experimental studies, helping to explain molecular dynamics and to state new hypotheses, most often about the structure of interactions. A relatively small amount of exp...

The JAK-STAT signaling network in the human B-cell: an extreme signaling pathway analysis.

Large-scale models of signaling networks are beginning to be reconstructed and corresponding analysis frameworks are being developed. Herein, a reconstruction of the JAK-STAT signaling system in the human B-cell is described and a scalable framework for its network analysis is presented. This approach is called extreme signaling pathway analysis and involves the description of network propertie...
